Job Description

Designation: Principal Investigator/Senior Principal InvestigatorDepartment: Discovery BiologyLead a team of scientists (10-15) on to deliver on complex molecular biology projects, including therapeutic antibody engineering (bispecifics, trispecifics etc), generation of display libraries (phage/yeast/mammalian), NGS-based technologies, therapeutic mRNA, and introduction of automation. Be the subject matter expert in these areas. Key Responsibilities:

• Independently perform HT gene cloning, construct designs for expression in E. coli, Pichia and mammalian cells.
• Design and execute mRNA in vitro transcription and high-volume plasmid DNA generation projects at lab scale.
• Antibody gene cloning from various sources- Eg., Hybridoma, B cells,
• Library generation-phage/yeast/mammalian display
• Engineering, designing and generation of Bi-specifics, Multi-specifics, CAR constructs etc.
• Engineered antibody expression and purification from various expression systems and QC by various methods
• Guide, inspire, motivate and train research scientists in the team at various levels
• Data interpretation and preparation of detailed reports for sharing with clients and attend telecons
• Provide technical and scientific input in project design and proposal writing
• Resource management for the molecular biology team
• Improve lab efficiency and streamline the operational processes to increase productivity
• Analyze operational problems, identify solutions, and implement corrective actions.
• Identify and implement development opportunities for the team members
• Contribution to vendor management, inventory keeping, general laboratory maintenance, quality and safety
• Compliance to Regulatory, Safety and Quality policies of Syngene for self and team
• Follow environment, health, and safety (EHS) requirements always in the workplace ensuring individual and lab/plant safety
• Attend training on environment, health, and safety (EHS) measures imparted company

Educational Qualification: Ph.D. (Biochemistry, Molecular Biology or related discipline) with 8-12 yearsTechnical/functional Skills:

• Relevant complex gene cloning experience in biotech, pharma or academia for expression in Pichia, E. coli and mammalian cells
• In vitro transcription of mRNA and plasmid DNA in mg scale.
• Relevant antibody-based research experience recombinant antibody generation, designing and generation of Bi-specifics, Multi-specifics, CAR-T molecules etc.
• Antibody gene cloning and sequencing from various sources- Eg., Hybridoma, B cells etc. and by NGS based technologies
• Antibody Library generation-phage/yeast/mammalian display
• Proven strong technical expertise in protein purification including complex antibodies from expression systems (Mammalian, E. coli, Insect cells and Pichia) using various chromatographic techniques
• Expertise in analytical techniques like SE-HPLC, BIAcore, Octet, etc
• Understanding of Mass spectrometry, protein sequencing etc.
• Knowledge of Basic bioinformatics tools-Handling of Molecular Biology analysis tools like Sequencher, Vector NTi, SnapGene, Geneious etc.
• Excellent verbal and written communication and interpersonal skills.
